Two separately labelled vials of white lyophilized powder at 10 mg each, and the two could hardly be further apart in scale. FOXO4-DRI is a 46-residue chain above 5300 daltons; Epithalon is a four-residue tetrapeptide near 390 daltons — roughly a fourteenfold difference in molecular weight at identical milligram fills. A 10 mg Epithalon vial therefore holds about fourteen times the molar quantity of the FOXO4-DRI vial beside it.

Store the sealed tray at -20°C (-4°F) for up to 36 months, with short-term holding at 2°C to 8°C (36°F to 46°F) acceptable and reconstituted material used within 14 days at 2°C to 8°C. The two behave very differently in solution. FOXO4-DRI's all-D backbone makes it resistant to proteolytic degradation — that resistance is the entire point of the retro-inverso design. Epithalon is an unprotected L-amino acid tetrapeptide with a high proportion of exposed backbone and is far more hydrolysis-prone, so reconstitute only what an experiment consumes.

The two components approach cellular ageing from opposite ends and share no target. FOXO4-DRI works by disrupting a protein-protein interaction: Baar and colleagues designed it in 2017 to interfere with FOXO4 binding to p53, and Bourgeois and colleagues showed in 2025 that the disordered p53 transactivation domain is the actual binding site. Displacing p53 from that interaction drives nuclear exclusion and selective apoptosis in senescent cells. Epithalon does the opposite kind of work entirely — it is a pineal tetrapeptide studied against telomerase upregulation and telomere length endpoints, a transcriptional and chromosomal question rather than a protein interaction one.
